1. Is coral reef being referred to in here this specific task, the same as the one over which we will have to fly the transect line? If it is the same one, do you expect to identify the points of interest as we move along from one black end of the coral reef to the other black end?
2. Is it acceptable to take a screenshot of the whole coral reef before or after the transecting is done and then identify those points of interest on the image?

The coral reef is the entire rectangle, with 27 squares, that you must fly the transect line over. You must then map the six points of interest in the coral reef - the six items will be in the squares of the coral reef. So yes, you are identifying and mapping the points of interest on the same coral reef you are flying a transect over.
Companies may identify points of interest at the same time they fly the transect line over the coral reef or companies may fly the transect to get points for that part, then go back and survey it again to create their map.
For EXPLORER companies, taking a screenshot of the coral reef would be considered manual mapping if it requires hitting a button/key on your control system / joystick / keyboard / etc. It would still be autonomous if the ROV pilot manuevers the ROV over the coral reef and without further manipulation, the map is created.

Thanks for the clarification on these questions. I had one regarding the manual vs autonomous distinction. It was a little unclear in this post (https://forums.marinetech2.org/viewtopi ... =29&t=1227) as to if taking a screenshot of the reef from above counted as autonomous. You said here that manually pressing capture will be considered manual.
Are we allowed to set our ROV into a mode for trying to find the reef and create a map? I'm hoping to avoid running semi-intensive computer vision the whole mission run.

You are allowed to set your ROV into a certain mode for part of the mission, and then have it in a different mode for another part of the mission. You do not need to run with enhanced computer vision the entire time.
